Justine Henin-Hardenne claimed the title, defeating home player Mary Pierce in the final, 6-1, 6-1 to win the Women's Singles tennis title at the 2005 French Open. This was Henin-Hardenne's second French Open, and her first in a string of three consecutive.

Anastasia Myskina was the defending champion, but lost in the first round to unseeded María Sánchez Lorenzo. Myskina became the first French Open champion to lose in the first round of her title defense and the only one to do so until Je?ena Ostapenko in 2018.

This was the first French Open main draw which future champion Ana Ivanovic competed in. In just her second Grand Slam main draw appearance (and first as a seeded player), she reached the quarter-finals, losing to Nadia Petrova after defeating hometown player Amélie Mauresmo in the third round.[1]
